Unzipping DNA : A hypothesis on correlation during replication

چکیده
Somendra M. Bhattacharjee Institute of Physics, Bhubaneswar 751 005, India [email protected] (February 1, 2008) Based on the result of an unzipping phase transition by a force in a model of DNA, we hypothesize that the DnaA-type proteins act as a pulling agent with a force slightly less than the critical force for unzipping. The dynamic (space-time) correlation of unzipping then drives the subsequent events of replication. In such a correlation driven scenario, there is no need of a replisome as a structural unit and this elusive replisome may not exist at all.

Unzipping DNA - towards the first step of replication
The opening of the Y-fork the first step of DNA replication is shown to be a critical phenomenon under an external force at one of its ends. From the results of an equivalent delocalization in a nonhermitian quantum-mechanics problem we show the different scaling behavior of unzipping and melting. The resultant long-range critical features within the unzipped part of Y might play a role in the ...
متن کاملTwo heads are better than one: regulation of DNA replication by hexameric helicases.
DNA replication is tightly regulated in a cell cycle to ensure the integrity of genomic information during successive passages. The replication process can be divided into three major steps as follows: the initial assembly of prereplication complex (pre-RC) at the replication origin, the distortion of the origin (or origin melting) for replication initiation, and the elongation phase during DNA...
متن کاملThe Boltzmann factor, DNA melting, and Brownian ratchets: Topics in an introductory physics sequence for biology and premedical students
Three, interrelated biologically-relevant examples of biased random walks are presented: (1) A model for DNA melting, modelled as DNA unzipping, which provides a way to illustrate the role of the Boltzmann factor in a venue well-known to biology and pre-medical students; (2) the activity of helicase motor proteins in unzipping double-stranded DNA, for example, at the replication fork, which is ...
متن کاملThermodynamic relations for DNA phase transitions
The force induced unzipping transition of a double stranded DNA is considered from a purely thermodynamic point of view. This analysis provides us with a set of relations that can be used to test microscopic theories and experiments. The thermodynamic approach is based on the hypothesis of impenetrability of the force in the zipped state. The melting and the unzipping transitions are considered...
متن کاملUnzipping dynamics of long DNAs.
The two strands of the DNA double helix can be "unzipped" by the application of approximately 15 pN force. We analyze the dynamics of unzipping and rezipping for the case where the molecule ends are separated and reapproached at constant velocity. For unzipping of 50-kilobase DNAs at less than about 1000 bases per second, thermal-equilibrium-based theory applies. However, for higher unzipping v...
